灯盏细辛胶囊治疗缺血性脑卒中的药理机制研究(1)
摘要 目的：利用网络药理学研究灯盏细辛胶囊治疗缺血性脑卒中的药理机制。方法：采用TCMSP等检索灯盏细辛胶囊的活性成分，并且得到其灯盏细辛胶囊活性成分对应的靶点;然后通过PharmGKB、Genecards2个数据库检索缺血性脑卒中相关靶点，使用METASCAPE软件对两者取交集的49个靶点进行GO注释分析和KEGG通路分析。采用Cytoscape 3.7.1软件建立活性成分-靶点-通路网络模型。结果：灯盏细辛胶囊可能通过补体和凝血级联通路、HIF-1信号通路、PI3K-Akt信号通路、FoxO信号通路、核因子-κB信号通路、脂肪细胞因子信号通路等多个通路治疗缺血性脑卒中。结论：灯盏细辛胶囊体现了中药多成分、多靶点、多通路的特点，为进一步阐释灯盏细辛胶囊治疗缺血性脑卒中的药理机制提供了理论依据。

Abstract Objective：To study the pharmacological mechanism of Dengzhan Xixin Capsule in the treatment of ischemic stroke by network pharmacology.Methods：The active components of Dengzhan Xixin Capsules were retrieved by TCMSP and the corresponding targets were obtained.Then，through PharmGKB and Genecards，the 2 databases were used to search for ischemic stroke-related targets，and METASCAPE software was used to perform GO annotation analysis and KEGG pathway analysis on the 49 targets where the 2 intersect.Cytoscape 3.7.1 software was used to establish the active ingredient-target-channel network model.Results：Dengzhan Xixin Capsules may treat ischemia through multiple pathways such as complement and coagulation cascade pathway，HIF-1 signaling pathway，PI3K-Akt signaling pathway，FoxO signaling pathway，NF-kappa B signaling pathway，and adipokine signaling pathway.Conclusion：Dengzhan Xixin Capsules embody the characteristics of multiple ingredients，multiple targets and multiple pathways of traditional Chinese medicine，which provides a theoretical basis for further elucidating the pharmacological mechanism of Dengzhan Xixin Capsules in the treatment of ischemic stroke

中药在治疗缺血性脑卒中(Cerebral Ischemic Stroke，CIS)方面具有多组分，多靶点，整体调节优势，已成为防治缺血性脑卒中的重要组成部分。灯盏细辛胶囊是由灯盏细辛组成。具有活血化瘀的作用。灯盏细辛胶囊的药理学机制包括降低血液黏稠度，抑制血小板聚集，抗缺血及灌注损伤，清除氧自由基、抗氧化，改善脑微循环及器官血流动力学等多方面作用等[1-2]。多靶点传统中药可能对缺血性脑卒中更有效。灯盏细辛胶囊的功效已经在临床实践中广泛应用，灯盏细辛胶囊治疗缺血性脑卒中的机制尚不清楚。
网络药理学作为一种系统的方法，能够帮助分析传统中药和疾病之间的药理数据。网络药理学将生物学特征与化学结构特征联系起来，并组建网络模型，有助于深入分析涉及药物药理作用的成分[3]。基于网络药理学的方法能帮助我们对中成药灯盏细辛胶囊所涵盖的多个层次作用进行分析。我们拟构建灯盏细辛胶囊-靶标-缺血性脑卒中的网络，对灯盏细辛胶囊多成分、多靶点、多途径的特征进行系统研究，并揭示中成药灯盏细辛胶囊治疗缺血性脑卒中的药理作用及其作用机制。
1 材料与方法
1.1 化学成分信息 通过中药系统药理学数据库和分析平台TCMSP(http：//lsp.nwu.edu.cn/index.php)[4]和PubChem Compound数据库(https：//www.ncbi.nlm.nih.gov/pccompound/)，分别检索灯盏細辛胶囊的化学成分。以口服生物利用度(OB)≥30%和类药性(DL)≥0.2为限定条件，筛选灯盏细辛胶囊的化学成分-靶点信息。CIS相关靶点的获从PharmGKB(https：//www.pharmgkb.org/)、Genecards(https：//www.genecards.org/)2个疾病相关靶点的数据库检索[5-6]。, http://www.100md.com(吴沣 彭金香)
